Introduction
For many years, the main treatment for depression has been antidepressant pills. These drugs, like SSRIs, work by changing the levels of chemicals in the brain. They have helped many people. But for others, they don’t work well or cause bad side effects.
This has led scientists to look for new ways to treat depression. They have started to study psychedelic drugs again. A new study in the journal Nature Medicine looked at a psychedelic called DMT. It found that a single dose could give fast and lasting relief to people with depression (1). Other research on mice shows that DMT may even repair the brain (2). This new science offers a whole new way to think about healing from depression.
How Does a Single Dose of DMT Affect Depression?
A recent study looked at the effects of a short-acting, lab-made version of DMT. The study was a Phase IIa trial. This means it was focused on safety and finding the right dose. The study included people with moderate to severe depression. Some people got a dose of DMT, while others got a placebo (a dose with no drug in it). The results were impressive.
People who got DMT had a big drop in their depression scores. These scores were measured with a tool called the MADRS (Montgomery-Åsberg Depression Rating Scale). How Does DMT Work in the Brain?
Scientists believe DMT works by helping the brain change and heal itself. This is called neuroplasticity. It’s the brain’s ability to make new connections between brain cells.
A 2026 study in the journal Translational Psychiatry showed how this might work. Scientists studied mice that were under a lot of stress. The stress caused the mice to show signs of depression. They lost interest in things they usually enjoyed. But a single dose of DMT changed that. The mice started acting like their old selves again (2).
The study found that DMT caused the growth of new brain cells. This is a process called neurogenesis. It happened in a part of the brain called the hippocampus. The hippocampus is important for learning, memory, and mood. Stress can damage this part of the brain. The DMT treatment seemed to repair that damage. Is DMT a Safe Treatment for Depression?
Safety is the most important thing when it comes to new treatments. This is especially true for a powerful drug like DMT. In the study, DMT was found to be mostly safe. The psychedelic experience, or “trip,” was strong but short. It only lasted about 20 to 30 minutes. This is much shorter than a psilocybin trip, which can last for hours. The side effects were mostly mild. They included things like headaches, feeling sick, and dizziness (1).
