While a healthy diet, exercise, and mental puzzles are standard recommendations, scientists are now exploring a surprisingly different approach: psychedelics. Could a substance famous for its role in 1960s counterculture actually hold the key to keeping our brains young and flexible?
A groundbreaking new study at the UC Berkeley Center for the Science of Psychedelics is setting out to answer exactly that. By investigating how psilocybin (the active compound in "magic mushrooms") affects the brains of healthy older adults, researchers are opening a new frontier in the science of aging. What is the PLASTICITY Study?
Launched in 2026, the PLASTICITY trial (Psychedelic Longitudinal Aging Study In Cognitively Healthy Older Adults) is the first psychedelic neuroimaging study specifically focused on healthy adults between the ages of 60 and 85 (1). Unlike most clinical trials that look for ways to treat existing conditions like severe depression or PTSD, this study is asking a different question: Can psilocybin promote positive outcomes and successful aging in people who are already healthy?
Participants in the study will receive a dose of synthetic psilocybin in a highly controlled, comfortable environment with trained guides. Before and after the experience, researchers will use advanced MRI scans to look for microscopic changes in the hippocampus—a part of the brain crucial for learning and memory (1) (2). They will also use psychological tests and surveys to measure changes in perception, emotional regulation, and feelings of social connection.
How Might Psilocybin "Reboot" the Brain?
To understand why scientists are so interested in psilocybin, we have to talk about "neuroplasticity." This is the brain's ability to reorganize itself by forming new neural connections. When we are young, our brains are highly plastic, allowing us to learn quickly and adapt to new situations. As we age, this plasticity naturally declines, making it harder to learn new things and sometimes leading to entrenched, negative thought patterns.
Previous studies in animals have shown that psilocybin can rapidly increase the number of synaptic connections in the brain, essentially resetting plasticity to a more youthful state (1) (2). What Do We Know About Older Adults and Psychedelics?
Historically, older adults have been largely left out of modern psychedelic research, making up only about 1.4% of participants in clinical trials (1). However, recent observational studies are beginning to shed light on how older adults respond to these substances.
A 2024 prospective cohort study followed older adults (aged 60 and above) who participated in guided psychedelic group retreats (3). The researchers found that the older adults experienced significant improvements in their overall well-being, similar to a matched group of younger adults. Interestingly, the older adults reported that the acute "intensity" of the psychedelic experience was lower for them than for the younger participants. For the older group, the improvements in well-being were not predicted by how intense the "trip" was, but rather by the deep sense of interpersonal connection and "togetherness" they felt during the group activities (3).This suggests that for older adults, the social and relational aspects of psychedelic therapy may be just as important—if not more so—than the pharmacological effects of the drug itself.
